I would suggest if you get to Ramstein, and nothing heading to Rota from there, then head to Frankfort/Hahn and fly to Jerez from there. Using Ryanair.

Plenty of European discount airlines fly the Germany/Spain route taking tourists to and from holiday destinations. For example, looking at Air Berlin website, they have a mid Sept one way fare from Stuttgart to Seville for about 120 Euros.....

Stuttgart to Rota would be rare. More likely Stuttgart to Sig as most of the traffic is heading to the African Continent via Sig.

We planned a trip to Europe this fall. Initially I was planning to fly from the East Coast to Rota and spend a week or so in Andalusia, and then hoping to get a flight to either Ramstein or Stuttgart to visit friends in Germany, and then fly back via Ramstein or Spangdahlem. Now we have to be early September in Germany; how difficult is it to fly from Germany to Rota? Does Stuttgart have any direct flights, or would one have to go via Sigonella to Rota?
